SEMAX

Semax is a synthetic heptapeptide (Met-Glu-His-Phe-Pro-Gly-Pro) derived from the 4–10 fragment of adrenocorticotropic hormone (ACTH), with the Pro-Gly-Pro C-terminal extension added to confer metabolic stability against rapid enzymatic degradation. Its primary mechanism relevant to cognitive and neuroprotective research involves rapid upregulation of brain-derived neurotrophic factor (BDNF) and its signaling receptor TrkB in the hippocampus, with studies reporting approximately 3-fold increases in exon III BDNF mRNA and concomitant TrkB phosphorylation following acute administration. Additional mechanisms under investigation include activation of dopaminergic and serotonergic systems, possible MC4R and MC5R interactions, and neuroprotective effects in models of ischemia, optic nerve damage, and cognitive impairment.
