Melanotan 2 is a cyclic synthetic analog of alpha-melanocyte-stimulating hormone with broad melanocortin receptor activity, demonstrating full agonist binding at MC1R, MC3R, MC4R, and MC5R with minimal affinity for the adrenocortical MC2R. Unlike the receptor-selective MT-1, its non-selective binding profile has made it a research tool for studying the distinct contributions of individual melanocortin receptor subtypes, particularly in the contexts of cutaneous pigmentation (MC1R), energy homeostasis (MC3R/MC4R), and centrally mediated behavioral research (MC4R). The MC4R-mediated effects observed in rodent and primate research models led directly to the clinical development of bremelanotide (PT-141), which received FDA approval for female hypoactive sexual desire disorder.
