Nicotinamide adenine dinucleotide (NAD+) is a fundamental coenzyme present in all living cells, functioning as an electron carrier in mitochondrial redox reactions central to ATP synthesis and as a required substrate for NAD+-dependent signaling enzymes that regulate cellular aging, stress response, and genomic integrity. Research has established that NAD+ levels decline progressively with chronological aging and are further reduced by obesity and sedentary conditions, impairing the activity of sirtuin deacylases (SIRT1–7), poly-ADP-ribose polymerases (PARPs), and CD38 ectoenzymes that govern DNA repair, mitochondrial biogenesis, and inflammatory regulation.
