SERMORELIN

Sermorelin is a synthetic 29-amino acid polypeptide representing the minimal fully bioactive N-terminal fragment (1–29) of endogenous human growth hormone-releasing hormone (GHRH), and was the first GHRH analog approved by the FDA for diagnostic assessment and treatment of growth hormone deficiency. It binds GHRH receptors on anterior pituitary somatotrophs to stimulate endogenous GH synthesis and pulsatile secretion, with the mechanistic distinction that its activity remains subject to negative feedback regulation by somatostatin, making supratherapeutic GH elevation physiologically self-limiting. Research has examined its utility in models of adult-onset GH insufficiency, somatotropic axis diagnostic testing, and age-associated decline in pituitary responsiveness.
