KPV

KPV (Lys-Pro-Val) is the C-terminal tripeptide of alpha-melanocyte-stimulating hormone (α-MSH) and the minimal sequence required to retain the parent neuropeptide’s anti-inflammatory activity. Unlike full-length α-MSH, KPV’s anti-inflammatory mechanism is not primarily mediated through melanocortin receptors but through PepT1, a di/tripeptide transporter upregulated in colonic epithelium during inflammatory conditions, facilitating direct intracellular uptake and subsequent inhibition of NF-κB-driven cytokine production. Murine models of intestinal inflammation have demonstrated significant reductions in inflammatory infiltrates, accelerated recovery, and attenuated mucosal damage following KPV exposure, establishing it as a compound of ongoing interest in gut inflammation and epithelial barrier research.
