CJC-1295 without DAC, also designated Modified GRF(1-29), is a tetrasubstituted synthetic analog of the 29-amino acid N-terminal fragment of growth hormone-releasing hormone (GHRH) engineered for enhanced resistance to dipeptidyl peptidase-IV (DPP-IV) cleavage while retaining a short plasma half-life of approximately 30 minutes. At the receptor level, it binds GHRH-R on anterior pituitary somatotrophs and activates the Gs/adenylyl cyclase/cAMP/PKA/CREB cascade to stimulate growth hormone synthesis and secretion in discrete, physiologically pulsatile patterns. Research has employed this analog to model the pulsatile GH secretory axis and study the downstream effects of preserved episodic GH and IGF-1 release in controlled experimental settings.
